Quadrant Streaking Virtual Lab
Description
The Quadrant Streaking Virtual Lab teaches students how to isolate bacterial colonies using aseptic technique and the four-quadrant streak plate method. Working at a realistic virtual bench, students flame the inoculating loop and aseptically transfer an inoculum from a culture tube to a nutrient agar plate. They then draw the streaks across all four quadrants by hand, flaming the loop and rotating the plate between areas. After incubation, students see colonies grow along their own streak lines and examine isolated colonies to describe their morphology. A troubleshooting section and a comprehensive final quiz complete the lab.
Learning Outcomes
By the end of this lab, students will be able to apply aseptic technique to transfer bacteria from a culture tube to an agar plate. They will perform the quadrant streak method correctly, flaming the loop and rotating the plate between areas, then label and incubate plates properly. Students will explain how streaking dilutes the inoculum to produce isolated colonies, each arising from a single cell, and how this is used to obtain pure cultures. They will also describe colony morphology and identify common streaking errors that prevent isolation.
