Endospore Stain Virtual Lab

Description

The Endospore Stain Virtual Lab lets students perform the Schaeffer–Fulton spore stain step by step, using real photographs of laboratory tools at a realistic virtual bench. Students prepare and heat-fix a smear of Bacillus megaterium. They then steam it with malachite green over a boiling water bath, keeping the stain from drying out, let the slide cool, rinse it, and counterstain with safranin. Under an oil-immersion microscope, they observe green endospores within pink vegetative cells and compare spore-forming species with different spore positions. The lab also includes a troubleshooting section on common technical errors and a comprehensive final quiz.

Learning Outcomes

By the end of this lab, students will be able to prepare and heat-fix a bacterial smear and perform the Schaeffer–Fulton endospore stain correctly. They will explain the role of heat, malachite green, water, and safranin, and how the resistant spore coat affects staining. Students will distinguish green endospores from pink vegetative cells under oil immersion and classify spore position as central, subterminal, or terminal. They will also identify common staining errors and relate endospores to sterilization and to diseases such as anthrax, tetanus, and botulism.
